项目摘要

The objectives of the present research are (1) validity evaluation of introduction of self-compatibility in breeding of common buckwheat, a typical self-incompatible species, by comparison of selection efficiency of rutin content between self-incompatible (SI) and self-compatible population (SC), and (2) QTL analysis for rutin content in common buckwheat by using high and low rutin content lines. The outline of research results are as follows ;(1) F4 and F5 population of SI and SC lines were cultivated and their seed rutin contents were quantified using HPLC. The individuals of 10% of higher ranks were selected in each population and their average rutin contents in next generation, namely F5 and F6, were investigated. Heritability of rutin contents were higher in self-compatible populations than in self-incompatible ones. These results indicate that introduction of self-compatibility into common buckwheat is valid for selection of other quantitative traits as well as rutin content.(2) Seed rutin contents in F1 and F2 plants of reciprocal cross between high and low rutin content lines were quantified. The F1 showed intermediate rutin content between high and low lines, but rutin content of F2 bulk seed was close to that of low content line. The high selection efficiency in self-compatible population indicates that additive effect is large in inheritance of rutin content. QTL analysis was not done because individual rutin content quantification in F2 plants was impossible due to insufficient seed yield for analysis.
本研究的目的是:(1)通过比较自交不亲和群体(SI)与自交亲和群体(SC)之间的芦丁含量选择效率,评价引入自交亲和育种在普通荞麦这一典型自交不亲和物种中的有效性;(2)利用高、低芦丁含量系对普通荞麦中芦丁含量进行QTL分析。研究成果概述如下:(1)培养SI系和SC系F4和F5群体,用高效液相色谱法测定其种子芦丁含量。在每一种群中选取10%的高阶个体,测定其下一代F5和F6的平均芦丁含量。自交亲和群体芦丁含量的遗传力高于自交亲和群体。上述结果表明,引入自亲和性对其他数量性状和芦丁含量的选择是有效的。(2)对高、低芦丁含量系正交F1、F2植株的种子芦丁含量进行了定量分析。F1的芦丁含量介于高、低含量系之间,而F2散装种子的芦丁含量接近低含量系。自交亲和群体选择效率高,说明芦丁含量遗传的加性效应大。由于种子产量不足,无法对F2植株进行单株芦丁含量定量分析,因此未进行QTL分析。
