We investigated to determine if implantation of bone marrow-derived cells can reconstruct smooth muscle layers in injured urinary bladder walls and restore function. Injury was achieved 3 days prior to implantation by applying a chilled iron bar to half of the bladder wall of female nude mice. Bone marrow cells were harvested from femurs of male mice and cultured for 7 days prior to implantation of dividing cells into the injured bladder walls. For controls, only the culture medium was injected. Before implantation and 14 days after, the urinary bladders were analyzed by immunohistochemistry, quantitative analysis of smooth muscle gene expression, and Cystometric investigation. All of the implanted urinary bladders had smooth muscle layer-like structures containing smooth muscle cells differentiated from bone marrow-derived cells. The implanted bladders expressed higher levels of smooth muscle mRNAs and improved detrusor functions 14 days after implantation compared to injured bladders prior to implantation. Only one control mouse survived beyond 8 days after injection of the control medium. After 14 days, it did not have any smooth muscle layers. These results was published in English journal.
