広汎性発達障害の病因は未だ明らかではない。本研究者らは、眼球運動と機能的脳画像を用いて脳機能障害部位について研究を行った。高機能自閉症、アスペルガー症候群の成人例13名と年齢をマッチさせた健常者13名に種々の眼球運動課題を行ってその結果を比較したところ、広汎性発達障害群(PDD)では、アンチサッカードの視標を見てしまうエラー、垂直sinusoidal smooth pursuitにおけるゲインの低下、step-ramp smooth pursuitの潜時の延長、アンチサッカード、記憶誘導性サッカードの速度の低下が認められた。また、垂直方向のsmooth pursuitでは上向き方向のpeak velocityの低下による非対称が認められた(上方向の最高速度:2.88°/s,下方向の量高速度:8.91°/s)。この結果は、未発達の状態を代償する小脳片葉-視床-前頭葉へのフィードバック経路の未熟性を示していると考えられる。広汎性発達障害の患者は、他者と視線を合わせないと言われ、社会における対人関係の障害の原因の一つとして他者の顔の表情の認知に異常があるという報告がある。そこで、未知の他者の喜怒哀楽の表情の写真を被験者にみせ、それが喜怒哀楽のうちのどれを表しているかを答えさせると同時に、賦活される脳部位を調べるために、GE社製SIGNA 1.5 Teslaの全身用MR装置を用いてGRE-EPI T_2強調画像をBOLD法により撮像した。得られた画像をSPM 5を用いて、喜怒哀楽の表情の信号からニュートラルな表情の信号を差し引くことにより、統計処理を行った。健常者8例の結果は、(A)happy:右後部帯状回、(B)angry:左前部島、(C)sad:右上側頭回の領域の賦活が認められた。現在PDD例で検討中である。
