项目摘要

ヒト腫瘍細胞と牛β-Caseinの共通抗原性を示す蛋白(BCLP)について解析するため、抗牛β-Caseinモノクローナル抗体を作成した。その中の12G2抗体を用いて子宮頸部腺癌細胞株のcDNA libraryをスクリーニングし、そのcDNA fragamentをクローニングした。その後、1998年にヒト脳細胞に存在するhippocalcinがクローニングされ、そのhippocalcinと我々がclonigしたBCLP fragamentの71-614までは99%の相同性を示し、それにはカルシウム結合能を有するEF-handをコードする部分を含むことが判明した。BCLPは、hippocalcinとは分子量が異なること、他の既知のカルシウム結合蛋白とは高い相同性を示さないため、カルシウム結合能を示すEF-handを持つ新しいカルシウム結合蛋白を我々は同定したと考えられる。また、子宮体部末分化癌の細胞株とその高運動能亜株を樹立し、その差異を検討しintegrin β1の発現がそれに関係していることを証明した。この細胞株には、Integrin β1と複合体を形成するCD9、erb2,3の発現が認められ、それぞれ運動能に関連していた。これらの物質の細胞内シグナル伝達にはカルシウム結合蛋白が関与している可能性が示唆されている。現在、BCLPにカルシウム結合能が実際に存在するか、さらにそれが細胞内シグナル伝達に関連しているか検討中である。血液中のBCLPのmRNAをRT-PCR法を用いて検出し、それが臨床応用可能か検討した。健常ボランテイアでは26例中4例15.4%陽性で、担癌患者では30例中19例63.3%陽性であった。これらより、Sensitivity63.65、Specificty86.4であり再発の早期診断など臨床的にゆうようではないかと推察された。

The common antigenic protein (BCLP) of bovine beta-casein in tumor cells was analyzed and anti-bovine beta-casein antibodies were prepared. The cDNA library of cervical adenocarcinoma cell lines was used to detect the presence of 12G2 antibodies. Since 1998, the existence of hipocalcin in human cells has been demonstrated to be 99% identical to that of its clonig and BCLP fragment No. 71-614, and the existence of an EF-hand in human cells has been identified. BCLP is a novel protein binding protein with different molecular weights and high affinity with other known proteins. This paper demonstrates the relationship between the development of integrin β1 and the differentiation of high motility cell lines in uterine somatic carcinoma. The expression of CD9 and erb2,3 in these cell lines is related to the expression of Integrin β1 complex. The possibility that the substance may bind to proteins in the intracellular system may be demonstrated. At present, BCLP can be combined with the actual existence of intracellular interaction. BCLP mRNA in blood was detected by RT-PCR, which may be used in clinical diagnosis. 4 of 26 healthy patients were positive in 15.4% and 19 of 30 cancer patients were positive in 63.3%.これらより、Sensitivity63.65、Specificty86.4であり再発の早期诊断など临床的にゆうようではないかと推察された。
