Investigation of the pathgenesis of SSPE by use of murine monoclonal antibodies against measles virus polypeptides.

用杂交瘤技术获得了抗麻疹病毒多肽的鼠源性单抗--抗HA69K、抗NP60K、抗F40K、抗M36K，这些抗体既能与裂解感染的麻疹病毒多肽反应，也能与持续感染的细胞内多肽反应。用这些抗体对2例麻疹巨细胞肺炎死亡患者的肺组织进行免疫荧光染色，并采集2例14岁女孩和8岁男孩的脑脊液标本，用电泳法、免疫学和病毒学方法进行分析。在这些脑脊液标本中检测到高滴度的麻疹特异性抗体和寡克隆的免疫球蛋白G条带，但用鼠源性单抗进行免疫荧光检测，未发现这些CSF的淋巴细胞中有病毒特异性多肽。

Murine monoclonal antibodies against measles virus polypeptides - anti HA69K, anti NP60K, anti F40K, anti M 36K - were obtained hybridoma technique which were specifically reacted with intracellular measles virus polypeptides not only by lytic infecton but also by persistent infection.Using these antibodies, lung tissues of two patients died with measles giant cell pneumonia were specifically stained by immunofluorescent technique.CSF samples of two patients with SSPE-14 year girl and 8 year boy-were obtained and were analyzed by electrophoretical, immunological and virological methods. High titer measles specific antibodies and oligoclonal IgG bands were detected in these CSF samples but, no virus specific polypeptides in lymphocytes of these CSFs were stained by immunofluorescence using murine monoclonal antibodies.
