Analysis of the Reproductive Failure by Using Monoclonal Antibodies Against Human Sperm

1.本文应用精子凝集试验（SAT）、精子制动试验（SIT）、酶联免疫吸附试验（ELISA）、免疫珠试验和混合抗球蛋白反应（MAR）对684例不孕妇女进行了抗精子抗体筛查。采用上述方法筛选出71例抗精子抗体阳性患者.用获能人精子免疫Balb/c小鼠脾细胞，获得抗人精子单克隆抗体。所有抗体对仓鼠卵-人精子穿透试验（ZSPT）均表现出抑制作用.与获能人精子特异性反应的单克隆抗体MH 61与Dynabeads M-450偶联。MH 61包被的微球与正常生育男性精子孵育后有明显的凝集反应，而与不育患者精子孵育后凝集反应较差。

Screening tests for antisperm antibodies were performed on 684 infertile women by Sperm Agglutination test (SAT), sperm immobilization test (SIT), enzyme-linked immunosorbent assay (ELISA), immunobeads test and mixed antiglobulin reaction (MAR). Seventy-one patients with circulating antisperm antibodies were selected by these methods.2. The monoclonal antibodies against human sperm were obtained by using Balb/c mouse spleen cells immunized with capacitated human sperm. All antibodies showed inhibitory effects on Zona free hamster egg-human sperm penetration test (ZSPT).3. Monoclonal antibody, MH61, which reacts specifically with capacitated human sperm was conjugated with Dynabeads M-450. A remarkable agglutination of MH61 coated beads was observed after incubation with sperm of fertile men, while poor agglutination activity was observed in sperm of infertile patients.
