【目的】緑茶カテキンの50%以上を占める(-)ーエピガロカテキンガレ-トはENNGによる発癌に対してプロモ-ション(P)を抑制する抗P作用があり、さらに移植した固形腸瘍の増殖を抑制する作用のあることが動物実験により確認されている。しかし、著者らの提唱している緑茶の直接摂取による、発癌のPおよびイニシエ-ション(I)に対する抑制作用については検討されていない。そこで、緑茶直接摂取のENNGによるマウスの十二指腸発癌に対する抗Pおよび抗I作用について検討を加えた。またウ-ロン茶および紅茶の抗P作用についても検討を行った。【方法】8週齢のC57BL/6NCrj雄マウスを用い、市販粉末飼料(オリエンタル酵母、飼育用MF粉末)に、セルロ-スを2%添加した対照群用飼料、同粉末飼料にそれぞれの茶粉末を2%添加したものを実験計画に沿って与えた。4週間のI期間はENNGの100ppm水溶液を自由摂取させた。その後の14週間にわたるP期間は水道水を自由摂取させた。飼育後解剖し、胃および十二指腸部分を摘出後、実態顕微鏡下で粘膜状態を観察し、腫瘍の発生個数、大きさ等を測定した。【結果】2回の実験の結果、対照群の腫瘍発生率は58、69%であった。P期間に緑茶飼料を与えた群の発生率は25、22%と有意に低かった。また、I期間に緑茶飼料を与えた群も、20%で有意に低かった。P期間にウ-ロン茶、紅茶飼料を与えた群の発生率は53、56%で対照群と有意差はなかった。以上のこと等から、緑茶の直接摂取はENNGによる十二指腸発癌に対して抗P、抗I作用のあることが確認できた。ウ-ロン茶の浸出液には、変異原性抑制作用が認められているが、本実験からは紅茶と共に、抗P作用は認められなかった。
