Cytogenetic and molecular genetic studies were carried out on the atomic-bomb survivors who had been exposed within 500 meters from the hypocenter.The main results obtained in the past three years :1.Follow up of the heavily exposed A-bomb survivorsAnnual follow up of the exposed people revealed six death, one each of acute myeloid leukemia, T cell malignant lymphoma and neoplasia of the kidney, and three of cerebral bleeding. Two survivors were found to have colon cancer (studied conducted in 1994,1995 and 1996 year)2.Establishment of methodology for biological dose estimation by FISHFluorescence in situ hybridization method which identify six painted chromosomes by two color was established. The detection rate of chromosome aberrations was almost the same as that of G-banding method, representing its utility for many exposed people.3.A high incidence of meningioma was found among heavily exposed groupAs two A-bomb survivors were found to have meningioma among 36 heavily exposed people, 38 meningioma patients in exposed group as well as 607 patients in non-exposed group were collected from the database of Hiroshima Tumor Registration. The group exposed more than one gray had significantly higher incidence of meningioma.4.A significant difference was observed in cytogenetic and molecular genetic findings between heavily exposed leukemia patients and non-exposedLeukemia patients exposed to more than one gray had complex chromosome aberration (p<0.05), less frequencies of leukemia-type specific chromosome translocation (p<0.005), RAS point mutation (p<0.07), MLL rearrangement (p<0.03), p53 mutation (p<0.07). These findings suggest different leukemogenesis process from that of de novo leukemia (studies conducted in 1994,1995 and 1996 year).
本文对原子弹爆炸后500米范围内的受照者进行了细胞遗传学和分子遗传学研究,三年来取得的主要结果:1.对原子弹爆炸后大量受照者的随访每年随访6例,其中急性髓细胞白血病、T细胞恶性淋巴瘤和肾肿瘤各1例,脑出血3例。1994年、1995年和1996年发现2例结肠癌患者。2. FISH生物剂量估算方法学的建立了荧光原位杂交法,用两种颜色标记6条染色体。染色体畸变的检出率与G显带法基本相同,说明G显带法对大部分受照者都有实用价值。3.高暴露组脑膜瘤的发病率高。从广岛肿瘤登记数据库中收集了暴露组38例脑膜瘤患者和非暴露组607例患者。高剂量暴露组与非高剂量暴露组的细胞遗传学和分子遗传学改变有显著性差异白血病类型特异性染色体易位(p <0.005)、RAS点突变(p <0.07)、MLL重排(p<0.03)、p53突变(p <0.07)的发生率明显低于对照组(p<0.05)。这些发现表明白血病的发生过程不同于原发性白血病(1994年、1995年和1996年进行的研究)。
