Y染色体特異配列の塩基配列決定による個人識別・父性診断について以下の検討を行った.1.DYZ1領域の反復配列Y10中,Kogan et al.の報告した3,511番〜3,659番の149bp断片を増幅するプライマーの一方の5′末端に-21M13配列を,他方の5′末端にM13reverse配列を付けたプライマーを用いてPCRを行った.男性試料からは目的とする増幅産物が得られた.女性試料ではほぼ同じ部位にスメア-が出現する場合があり,Y特異性が必ずしも高くないことが示唆された.複数の男性試料で得られた増幅産物のDye terminator法によるdirect sequencingを行ったが,現在のところ塩基置換や欠失等は確認されていない.2.DYZ1領域の反復配列Y10中,Nakahori et al.の報告した1,123番から2,146番までの1.024bp断片を増幅するプライマーの一方の5′末端に-21M13配列を,他方の5′末端にM13reverse配列を付けたプライマーを用いてPCRを行った.男性試料からのみ目的とする増幅断片が得られY特異性が高いと考えられた.増幅産物をアガロース電気泳動後切り出して,精製を行い,Dye terminator法によるdirect sequencingを行った.増幅断片長が1kbを越えるため,1回のcycle sequencingで全配列を決定することができず,この点について検討を行っている.3.Y染色体のマイクロサテライトY-29H39(DYS19)について検討を行ったところ5種類の対立遺伝子が検出され,父性遺伝も確認された.検出された対立遺伝子についてsequencingを行っている.
