本年度は、子宮平滑筋肉腫において9種類の腫瘍抑制遺伝子のLOHとMSIの有無を正常子宮平滑筋と比較することによって、子宮平滑筋肉腫の腫瘍性格を明らかにすることを目的とした。5例の子宮筋腫及び10例の子宮平滑筋肉腫とそれらに伴う正常子宮平滑筋層のパラフン包埋切片からそれぞれのDNAを抽出し、9種類の腫瘍抑制遺伝子の蛍光プライマーを用いてPCR法で増幅、日立自動DNAシーケンサーを用いて、9種類の腫瘍抑制遺伝子のLOH及びMSIの有無を解析した。その結果、5例の子宮筋腫では9種類の腫瘍抑制遺伝子のMSIは全く検出されなかった。一方、10例の子宮平滑筋肉腫は全例に少なくとも9種類の腫瘍抑制遺伝子うち1個所以上のMSIもしくはLOHが検出された。特にD14S267,p16,RBIにおいてはMSIが40%認められ、その他の領域では10-20%のMSI或いはLOHが認められた。MSIが頻度の高い陽性症例と臨床病理学的所見との相関を見ると、MSI陽性子宮平滑筋肉腫では核分裂が多く、予後が悪いことが判明した。これらの結果より、子宮平滑筋肉腫に少なくとも1個所以上のMSIもしくはLOHが検出され、またMSIあるいはLOHが高頻度の症例では細胞の核分裂が多いなど、相関が認められたことより、腫瘍抑制遺伝子のMSI及びLOHは子宮平滑筋肉腫の発生や増大に関与する可能が推測された。更に、これらの腫瘍抑制遺伝子のMSI及びLOHは子宮平滑筋肉腫の診断に使える可能性が示唆された。

This year, 9 types of uterine smooth muscle tumors were inhibited by LOH, MSI and normal uterine smooth muscle tumors. DNA was extracted from 5 cases of uterine fibroids and 10 cases of uterine fibroids with normal uterine fibroids, and amplified by PCR, Hitachi automated DNA analysis, LOH and MSI analysis of 9 types of tumor suppressor genes were performed. The results showed that 5 cases of uterine tendinous tumors and 9 kinds of tumor inhibition genes were detected by MSI. One side, 10 cases of uterine smooth muscle swelling, 9 types of tumor inhibition, 1 case of MSI, 2 cases of uterine smooth muscle swelling, 3 cases of uterine muscle swelling, 3 cases, 3 cases of uterine smooth muscle swelling, 3 cases, 3 cases of uterine smooth muscle swelling, 3 cases, 3 cases, 4 cases, 5 cases, D14S267,p16,RBI, MSI, MSI The frequency of MSI was high, and the correlation between clinical pathology findings and MSI positive uterine smooth muscle mass was high. As a result, there were fewer than 1 case of uterine smooth muscle mass, so MSI was not detected in LOH, MSI was not detected in LOH, and high frequency of cases, cell mitosis was more frequent, correlation was recognized, MSI and LOH of tumor inhibition gene were probably related to the development and enlargement of uterine smooth muscle mass. In addition, MSI and LOH of tumor suppressor genes may indicate the possibility of diagnosis of uterine smooth muscle tumors.
