We investigated neural cell migration through the rostral migratory stream (RMS) in the rat forebrain and obtained the new findings as follows.(A) Neurogenesis is ongoing in a small population of cells within the RMS during migration. Calretinin-immunoreactive neurons of the RMS co-express nestin (20.8%) and polysialylated neural cell adhesion molecule (61.8%). Calretinin-immunoreactive neurons in the RMS still express immature neural and neuronal phenotypes.(B) By the BrdU tracing method, newly formed periglomerular cells in the olfactory bulb are mostly replaced during life.(C) With the combined method of autoradiography using ^3H-thymidine and immunohistochemistry for a neuronal maker, 65.3-76.9% of bulbar neurons, following migration through the forebrain migratory route (RMS), undergo neuronal cell death during six weeks.(D) By preventing neural cell migration, we demonstrated neurogenesis by local neural stem cells in the olfactory bulb. Further, we revealed differential neurogenesis and gliogenesis by local and migrating neural stem cells in the brain.
