Die embryonale Mortalität ist eine wesentliche Ursache für mangelnde Fruchtbarkeit beimRind. Das Hauptziel unseres Projekts ist die Analyse des Transkriptomprofils von Embryonenund des Endometriums während des ganz frühen Trächtigkeitsstadiums in Bezug auf dieInitiierung einer Trächtigkeit nach Transfer von in vivo oder in vitro erzeugten Blastozysten.Biopsierte Embryonen werden am Tag 7 nach dem Östrus transferiert. Bis zum 24. Tagwerden Plasmaprogesteronwerte gemessen und über den Anstieg des Progesteronprofilswerden die Rezipienten in trächtige und nicht trächtige gruppiert. Sechs Tiere jeder Gruppewerden am Tag 16 geschlachtet und Embryo- und Endometriumproben gesammelt. WeitereTiere mit starkem Progesteronanstieg werden am 28. Tag geschlachtet und dieentsprechenden Proben gesammelt. Globale Transkriptomanalysen (Boviner Affymetrix Array)werden an den Blastozysten und Endometriumbiopsien in Abhängigkeit der dazugehörigen invivo Embryoentwicklung durchgeführt. So können Expressionsprofile von Embryonen am Tag7, 16 und 28 ermittelt werden. Vergleichende Analysen der Expressionsprofile zwischen denunterschiedlichen Stadien werden zur Identifizierung wichtiger Gene für die frühembryonaleEntwicklung führen und charakteristische Pathways zur Etablierung und Aufrechterhaltung derfrühen Trächtigkeit werden identifiziert werden. Darüber hinaus werden bei ausgewähltenGenen funktionelle Silencing Experimente durchgeführt, um ihre Bedeutung für dieEntwicklung näher zu untersuchen. Zusätzlich werden mittels vergleichender SequenzierungSNPs in den Kandidatengenen identifiziert, die mit Spermienparametern des Bullen und derNon-Return-Rate assoziiert werden.Embryonic mortality accounts for the largest proportion of reproductive wastage in cattlebreeding. The main goal of this project is to determine the transcriptional profile of bovineembryos and endoemtrium at early stage of development in relation to pregnancy successafter transfer of in vitro- or vivo-derived blastocysts. For this biopsied embryos will transferredto synchronized recipients on day 7 after estrus and until day 24 blood plasma concentrationswill be measured and pregnancy status of the recipients will be determined and animals will becategorized to pregnant (with rapid increase in plasma level) and nonpregnant (slow increasein plasma level). Six animals from each group will be slaughtered at day 16 and embryo andendometrium samples will be collected for analysis. Animals with rapid increase in plasmaprogestron concentration are further followed up and ultrasound guided pregnancy control willbe done on day 28. Six animals from pregnant and six animals from nonpregnant animals willbe subjected to slaughter and embryo samples will be collected for further analysis.Transcriptional profile of blastocyst biopsies collected perior to transfer will be performedbased on the pregnancy success after transfer. Moreover, comparative transcriptional analysisof in vitro versus in vivo derived embryos at days 16 and 28 of gestation will be determinedusing bovine Affymetrix array (~22,000 ESTs). The endometrial biopsies obtained prior totransfer will also be used for transcriptional and proteomic analysis based on the pregnancysuccess after transfer. With this it is possible to draw the expression profile of embryos at thetime of transfer (using blastocyst biopsies collected) and during the course of pregnancy(day16 and 28). Comparative anaylsis of these expression profiles will enable to identifycandidate genes and draw pathways related to embryo survival after transfer to recipients.Moreover, the functional role of some of the candidate genes in early embryonic developmentwill be investigated using RNA interefence and polymorphism of the candidate genes at singlenucleotide level will be determined to associate with the fertility trait of the bull i.e. the NonReturn Rate (NRR).
