项目摘要

HDLアポタンパクの分解は健常人であっても低い割合で起こっていると考えられる。さらに、HDLアポタンパクの分解速度が比較的速い人では低HDL値を示し、動脈硬化性の血管病変を起こしやすいと推測される。まず、本研究の目的と研究方法及び個人情報の保護方法について説明したのち同意を得た健常人の血漿をグラジエントポリアクリルアミドゲル電気泳動(NativeグラジェントPAGE)し、HDLアポタンパクA-1及びA-2に対するポリクローナル抗体を用いて、ウェスタンブロットを行ったところ、HDLとされている範囲より遥かに移動度が大きく粒子径の小さな位置にこれらの抗体と反応するタンパクのバンドが複数検出された。これらはすべて、これまでに報告のない新しいバンドであった。これらのバンドは銀染色及びCBBによるタンパク染色では染色性が悪く、脂質を染色するズダンブラックでも検出されなかった。次に、タンパクの良い分離が得られる等電点/SDS二次元電気泳動を行った。その結果、HDLのapoA-Iに相当する28kDaの位置にCBBでpI 5.2-6.0の範囲に、銀染色ではpI 4.6-6.2の範囲にスポットが検出された。さらに、apoA-1のポリクローナル抗体を用いたウェスタンプロットを行った結果、28kDaのスポットは抗apoA-I抗体と反応し、銀染色よりさらに広くpI 4.3-6.8の範囲で検出された。従って、apoA-Iには広い範囲の等電点を示すisoproteinが存在することが明らかになった。また、いずれの検出法においてもpI 5.7に大きなスポットが見られたが、特にウェスタンブロットでこの位置に分子サイズの異なるスポットが複数観察され、これらはapoA-I分解物と考えられた。以上より、血中低HDL値との相関は見られないものの、健常人においてもHDLの分解が起こっていることが明らかになった。

The correlation between HDL and serum levels was observed in healthy individuals.
