项目摘要

本邦において多くのHelicobacter pyloriが持つcag pathogenicity islandは多数のタンパクをコードする遺伝子群であり、それらのタンパクはTypeIV secretion systemを形成し、H.pyloriの意上皮細胞接着後にCagA蛋白を胃上皮細胞が胃上皮細胞内に送り込み、ケモカイン産生を刺激することが知られている。感染胃粘膜における菌側の因子を検討するため生検組織から分離培養されたH.pyloriのcag pathogenicity islandの遺伝子型について、cagA,cagEおよびVirD4の3つの遺伝子についての解析を中心に行った。また、宿主側がCagA蛋白をどの程度認識しているのか、これらの遺伝子の有無との関連を検討した。以上のことから得られた知見を論文で公表した。また、H.pyloriの胃粘膜上皮細胞への接着に重要な役割を果たすSabAをコードする遺伝子sabAについて組織学的胃炎、抗LewisX抗体との関わりについて検討し、13^<th> United European Gastroenterology Weekにおいて「Association between bacterial sabA status and presence of anti-Lewis X antibody in patients with Helicobacter pylori infection」として、ワークショップで発表した。

In this study, many Helicobacter pylori strains were found in cag pathogenicity island, and most of them were found in TypeIV secretion system. H.pylori was found in gastric epithelial cells, and then CagA protein was found in gastric epithelial cells.
