To recover fish habitat and restore rich biota in rural areas, Japan, genetic network and spatial structure among fish populations in agricultural ranals were investigated with mitochondrial and microsatellite DNA markets in this project. The main results were as follows.First, a phylogenetic analysis of the cytochrome b gene sequences in mitochondrial DNA was conducted to elucidate genetic variation and diversity of the loach Misgunius anguillicaudatus population in Japan. There were 154 haplotypes that were identified from 457 specimens collected from 124 sites. The phylogenetic tree based on the maximum parsimony method indicated four clades that inferred cryptic populations in Japan.Second, genetic structure of the loach populations in drainage mnals including main streams were analyzed using eight microsatellite DNA markers in the Shitada River basin composed of Yatsu paddy fields, Chiba Prefecture. The loach populations were estimated to be random mating, which conformed to the H … More ardy-Weinberg equilibrium. Genetic differentiation was confirmed significantly for a portion of the pairs of the populations among canals and the main stream.Third, selection of microsatellite DNA locus and preliminary analysis using the selected loci were conducted to clarify genetic properties of the Japanese eight-barbel loach Lefua echignnia populations in the upper basin of the Kokai River. A total of 12 loci were usable for the genetic analysis and a preliminary analysis by these selected loci showed that all populations had similar levels of genetic diversity. Genetic properties of the sampled populations appeared to differ between the Kokai and Ara-kawa river basins.Fourth, polymorphic microsatellite DNA markers were isolated in the Omono type of ninespine stidclebadc, genus Pungitius (GenBank accession nos. AB300827-AB300851), the Japanese eight-barbel loach (AB286032-AB286048) and the field gudgeon, Gnatbopogon elongatus elongates (AB264587-AB264600).
