加齢に伴い弾性線維が劣化すると、動脈は硬化・蛇行し、肺は肺気腫となり、皮膚は弛む。今後ますます進む高齢化社会において弾性線維の維持・劣化予防は重要な意義を持つと考えられる。われわれは、これまでに分泌タンパクDANCE(Developmental Arteries and Neural Crest EGF-like、またはfibulin-5)が弾性線維形成に必須であり、弾性線維構成分子をオーガナイズする分子機能を持つことを示してきた。また加齢にともない切断をうけたDANCEタンパクをより多く検出できることから、プロテアーゼ切断(プロセッシング)によるDANCEの機能変化が、弾性線維の形成・維持に関係しているという仮説をたてた。本研究は、弾性線維形成・維持におけるDANCEのプロセッシングが持つ意味を明らかにすることを目的とした。この目的のために、はじめからアミノ末端が切断されている切断型DANCE、切断箇所のアミノ酸配列を変えて切断されないようにした非切断型DANCEノックインマウスを作成した。現在、ヘテロノックインマウスを得ており、これをかけあわせることにより、今後、ホモノックインマウスを作成し、弾性線維の形成にDANCEプロセッシングがどのような役割を果たしているかを解析する予定である。また無血清培養の弾性線維構成系を用いて、切断型DANCEタンパクが弾性線維のコアとなるミクロフィブリル線維の上に沈着できないことを見いだした。以上から、加齢にともなう過剰なプロテアーゼによりDANCEは切断をうけ、弾性線維を維持できずに劣化するという仮説が考えられる。
