後天性表皮水庖症の原因蛋白であるVII型コラーゲン蛋白を抗原とした酵素結合免疫吸着定量法(ELISA)を作成するためには、精製したVII型コラーゲン蛋白が大量に必要となる。そのためにまずVII型コラーゲン蛋白を恒常的に発現する293A細胞を回転培養器により大量培養を行った。培養液の上清を硫安塩析し、透析した。次にVII型コラーゲン蛋白のNC-1domainに対する抗体であるLH7.2抗体を担体プロテインGに結合させたLH7.2抗体カラムを作成し、透析したサンプルを通したのちトリフルオロ酢酸にて抗体と蛋白の結合を切断しVII型コラーゲン蛋白を溶離した。しかしこの方法ではVII型コラーゲン蛋白以外の不純蛋白が混入してしまうことが判明したため、精製法をイオン交換アフィニティーに変更した。これで得られた290kDaの蛋白を選択し、それをさらにゲル濾過アフィニティーにて精製した。しかしこれは濃度が薄いため、アミコンを用いて濃縮した。しかし、大量精製用の1.2リットルのボトルの培養液から透析後に得られる蛋白量は23mgでそれがゲル濾過アフィニティー後は0.3mgとなり、濃縮後はさらにその半分の0.15mgで、回収率は0.65%である。ELISAの抗原として使用するためのVII型コラーゲン蛋白量が不足しているため、今後は回収率をあげるか、または培養器の数を増やすことが必要と考えている。そして十分量のVII型コラーゲン蛋白が準備できしだい、ELISAの作成を行なう予定である。

Type VII protein is the causative protein of acquired epidermal edema. It is necessary to prepare and purify type VII protein by enzyme-linked immunosorbent assay (ELISA). 293A cells were cultured in vitro in large quantities. The supernatant of the culture solution was analyzed and dialyzed. The NC-1 domain of the type VII protein binds to the LH7.2 antibody, and the LH7.2 antibody binds to the LH7.2 antibody. The LH7.2 antibody binds to the LH7.2 antibody, and the LH7.2 antibody binds to the LH7.2 antibody. The LH7.2 antibody binds to the LH7.2 antibody. This method has changed from the identification of the mixing of impure proteins other than type VII kuragonin protein to the exchange of purification methods. The 290kDa protein was selected and purified. The concentration of these substances is very high. 1.2 mg of protein was obtained after dialysis, 0.3 mg of protein was obtained after filtration, 0.15 mg of protein was obtained after concentration, and recovery rate was 0.65%. The amount of type VII protein in the ELISA antigen is insufficient, and the recovery rate in the future is increased. The preparation of a very high level of type VII protein and the preparation of an ELISA are predetermined.
