项目摘要

It is thought that the cancer of oral cavity is still feared as a fatal disease ease regardless of remarkable advancement of the diagnosis and the treatment method of the cancer of oral cavity and a big obstacle is given from the change to a morbid state of the face it to QOL. Little research of the relation between the cancer of oral cavity and the Wnt cell signaling route is advanced though β-Catenin is thought to be one of the oncogenes. It was thought that it was necessary to promote this research in the emergency for overcoming at the early stage of the cancer of oral cavity, examined as follows, and it reaped the benefit.1. Examination of irregular accumulation to cytoplasm and nucleus of B-Catenin of culture mouth cancer cell.Both what β-Catenin has accumulated abnormally in the cytoplasm and the nucleus and the thing that had not accumulated the label by the fluorescent antibody to recognize β-Catenin of the culture flatness cuticle cancer cell and the culture saliva glandular carcinoma cell, the observation with the electron microscope, and the retrieval whether β-Catenin had accumulated abnormally in the cytoplasm and the nucleus existed.2. Examination of gene mutation of β-Catenin of culture mouth cancer cell. RNA of β-Catenin of the culture flatness cuticle cancer cell and the culture saliva glandular carcinoma cell was extracted, the presence of the gene mutation was retrieved by the PCR-SSCP method, direct sequence method Denco mutation was tried in addition, and the decision of the position was tried. In the culture mouth cancer cell that had abnormally accumulated β-Catenin in the cytoplasm and the nucleus, the one with the gene mutation of β-Catenin and the one that not was existed. Moreover, the area was a part phosphorylated by GSK3.
