肝臓を生体外で再構築ためには,増殖・分化ポテンシャルの高い細胞を用い,3次元構造を形成させることで肝機能を発揮させる必要がある。そこで本研究では,(1)増殖・分化ポテンシャルの高い細胞として,小型肝細胞と骨髄間葉系幹細胞との共培養,(2)3次元構造を形成させるための細胞外環境の付与,を組み合わせることで,立体的なミニ肝組織の構築及びその形成機序の解明を目的とした。本研究により,患者の肝組織より小型肝細胞を分離培養し,同患者から採取した骨髄間葉系幹細胞を共培養,増殖させることで細胞数の確保とその増幅に限界があった小型肝細胞の臨床応用の可能性が高まると考えられる。
In vitro reconstruction of liver tissue, proliferation, differentiation and development of high level cells are essential for the development of liver function. The aim of this study is to: (1) co-culture of small hepatocytes and mesenchymal stem cells from bone marrow;(2) assignment of the outer cell environment to the formation of three-dimensional structure; and (3) elucidation of the three-dimensional liver tissue architecture and its formation mechanism. In this study, small hepatocytes from liver tissue of patients were isolated and cultured together with mesenchymal stem cells from bone marrow of patients, and the number of cells was increased to ensure the high possibility of clinical use of small hepatocytes.
