Durch Hormone oder Wachstumsfaktoren, die an spezifische Partner auf der Zelloberfläche binden, erhalten Zellen wichtige Informationen. Die Übermittlung dieser Informationen von der Zelloberfläche ins Zellinnere und an den Zellkern findet durch Wechselwirkung und chemische Veränderung von Proteinen statt. Proteine, die keine eigene katalytische Aktivität besitzen, können als Anker, Adaptor oder Plattform dienen und so weitere Proteine zusammenbringen. Gab1, Gab2 und das bei der Sequenzierung des Maus- und Menschengenoms entdeckte Gab3 sind Mitglieder einer Familie von Adaptorproteinen. Die biologische Funktion von Gab3 ist noch ungeklärt. Da die Milz und das Knochenmark viel Gab3 zu produzieren scheinen, könnte Gab3 eine Rolle bei der Blutbildung spielen. In diesem Projekt soll mit Hilfe biochemischer und molekulargenetischer Methoden die Funktion von Gab3 untersucht werden. Kernpunkt ist dabei die Herstellung von Mäusen mit inaktiviertem Gab3-Gen und ihre Charakterisierung. Hormones and growth factors that bind to specific partners on the cell surface convey important information to the cell. The transfer of this information from the cell surface to the inside of the cell and its nucleus involves the interaction and chemical modification of proteins. Proteins without catalytic activity can serve as anchor, adaptor, or scaffold to connect other proteins. Gab1, Gab2, and the new Gab3 - recently discovered after the sequencing of the mouse and human genomes - are members of a family of adaptor proteins. The biological function of Gab3 is not known. Since the spleen and the bone marrow seem to produce Gab3 in high amounts, Gab3 could play a role in the production of blood cells. This project aims to investigate the biological function of Gab3 using biochemical and moleculargenetic methods. A focal point is the generation and characterization of mice with an inactivated Gab3 gene.
