Neuronal circuits are formed by axons that have elongated and reached their targets. The tip of each axon, called the growth cone, explores extracellular guidance cues and can turn in the appropriated direction. In this project, we found that a Ca^<2+> nanodomain arising from an endoplasmic reticulum Ca^<2+> channel drives attractive guidance and that Myosin Va is released from the Ca^<2+> channel upon sensing the ^<2+> nanodomain leading to downstream signaling. We also showed that the dissociation of Myosin Va is sufficient for attractive growth cone turning.
