Analysis of antigens for proliferating T cells in patients with systemic lupus erythematosus.

Many lines of studies suggested that T cells play pivotal roles in the pathogenesis of systemic lupus erythematosus (SLE). We reported oligoclonal T cell proliferation in peripheral blood of patients with the disease previously. To identify the antigen peptides for the accumulated T cell clonotypes, we developed a T cell receptor cloning method from a single cell for the reconstruction of TCR molecules. Further, we successfully expressed HLA class I molecules with beta-2-microglobulin on the filamentous phages as a single chain for constructing an antigen peptide library. Screening of the library with the reconstructed TCR would determine candidate antigens for the proliferating T cells in SLE.
