This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Electrical transfer between the myocytes within the heart occurs at the gap junctions. Two connexons in juxtaposed membranes link to form a gap junction. An assembly of six membrane spanning proteins called connexins from a connexon. Cardiac cells express various isoforms of connexins which have distinct biophysical properties. In order to study patterns of myocyte-myocyte connectivity, the aim of this project is to study the heterogeneity of distribution of gap junctional proteins such as connexin43 in the mouse heart. Measures of heterogeneity of distribution include computing regional variations within the heart and computing anisotropy of distribution about the cell. Since the gap junction plaques are only a few microns in dimension, we would like to use a confocal microscope to obtain the high-resolution images. Since a mouse heart is approximately 7mm in diameter and 1 cm long the tiled images will need to be montaged to obtain the full image.
