This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. We hypothesize that participants with high levels of insulin resistance (IR), as determined by response to glucose tolerance testing (GTT), will show greater cognitive deficits. Further, we hypothesize that participants with IR will demonstrate cognitive improvement after treatment with Metformin, an insulin-sensitizing medication. Endpoints: Our first hypothesis, that participants with high levels of IR, as determined by response to GTT, will show greater cognitive deficits, will be answered by correlating GTT data with neurocognitive data. Our second hypothesis, that participants with IR will demonstrate cognitive improvement after treatment with an insulin-sensitizing medication, will be answered by testing for changes in neurocognitive data after treatment with Metformin. Further, we will test for changes in IR after treatment with Metformin.
