The SCID mouse: The skin xenograft core has developed methods to routinely generate severe combined
immunodeficient (SCID) beige (bg) double mutant mice engrafted with human skin in the form of split thickness
skin grafts or with synthetic skin constructs incorporating human skin cells. The three specific aims of the
core are (1) to provide members of the YSDRC (both established and junior investigators, and especially
investigators with YSDRC P/F projects) with limited numbers of C.B-17 SCID/beige mice engrafted with
healthy adult human skin or skin constructs for establishing the utility of these models for specific skin
disease-related studies; (2) to train members of the YSDRC to generate such mice once feasibility has been
established; and (3) to develop variant models of this technique for new and novel applications, specifically
focusing on synthetic skin equivalents constructed from distinct populations of cells both from patients with
specific skin diseases and/or that may be genetically modified to alter biologic functions. The core will also
continue evaluating alternative mouse recipient strains and methods to combine human skin grafts with
elements of the human hematopoietic system to better model inflammatory skin disorders and responses to
skin infections.
