This subproject is one of many research subprojects utilizing the
resources provided by a Center grant funded by NIH/NCRR. The subproject and
investigator (PI) may have received primary funding from another NIH source,
and thus could be represented in other CRISP entries. The institution listed is
for the Center, which is not necessarily the institution for the investigator.
Proteasomes regulate many facets of cellular metabolism. They are activated by complexes that bind to the ends of their cylindrical structures. We have previously reported (in Nature) the structure of a proteasome-activator complex. We are now working on a very different activator complex. Preparation of material is challenging, but we now have some preliminary crystals that diffract only very weakly on a home source. Our current goal is to collect native data that allow preliminary structure determination. This would be an important step on the path to an eventual high resolution structure of this 1.2 MDa complex.
