This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. We are developing a Fourier domain phase microscope (FDPM) for reflection-mode studies of cell electromotility. The FDPM uses a broadband light source together with a common-path phase-referenced detection scheme to allow depth resolved measurements of cell displacements at kHz bandwidths with angstrom-scale sensitivity. The low-noise and high sensitivity of SDPM will make it possible to measure the cell response to a unit impulse stimulus. Two-dimensional imaging can also be performed by preferably scanning the sample beam.
