项目摘要

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Background: Muscle loss after hip surgery requires a longer period of rehabilitation and delays the return to work. Hypothesis: Reducing the body's stress and increasing its ability to make protein will decrease muscle loss after hip surgery and help rehabilitation. Specific Aims and Procedures (summary): We will study how the body and muscle make protein after hip surgery and before discharge from the hospital. Further, we will test the effects of a nutritional supplement and a common drug on these processes. Experimental Design (summary): This study will have 5 groups of randomly assigned patients. Group 1 will serve as a normal control group and be studied only once before surgery. Groups 2-5 will be studied immediately after hip arthroplasty (during surgical recovery) and again prior to discharge from the hospital after surgical intervention. These groups include placebo (Group 2), amino acid (AA) supplementation (Group 3), ketoconazole administration (Group 4), and a combined group to receive both ketoconazole and AA (Group 5). Significance (summary): Reducing the loss of muscle after surgery will help with recovery and return to normal function.
