This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. OBJECTIVE: The long-range vision of this proposal is to develop imaging techniques to study mechanisms of action of therapeutic interventions in brain disorders. The immediate goal of this proposal is to study mechanisms of action of treatment in Idiopathic Parkinson's Disease. (IPD). In particular, we propose to study Lee Silverman Voice Treatment (LSVT). The mechanism of action of behavioral therapies, in general, and of speech therapy, is largely unknown. The specific project will use Parkinson's Disease (PD) as a model of brain disorders. The investigators will use speech therapy (Lee Silverman Voice Treatment, LSVT) as their treatment. The overall aim of this study is to use functional imaging to explore mechanisms of action of LSVT in the treatment of hypophonia in PD. METHODS: Subjects with PD will be recruited, imaged, trained in LSVT, and re-imaged. Four imaging techniques will be used: PET, functional MRI, transcranial magnetic stimulation, and resting state MRI.
