This study is an exploratory analysis of the difference in the prevalence of the JC Virus in oropharyngeal secretions and blood of healthy individuals, persons with Acquired Immune Deficiency Syndroms (AIDS), and pes with progressive multifocal leukoencephalopathy (PML). Saliva and throat washings as well as whole blood samples will be taken from each group. Saliva samples and throat washings will be analyzed for JCV using polymerase chain reaction (PCR). Whole blood samples will be collected and analyzed for JCV using PCR and for the presence of JCV in peripheral blood leukocytes (PBLs) using CD4 lymphocyte counts. These measures will be used to determine whether JCV is harbored in the oropharynx, whether a correlation exists between JCV levels in the oropharynx and those in PBLs and to determine whether the level and strain of JCV shed from the oropharynx correlates with the degree of immunosuppression. It is predicted that JCV will be present in oropharyngeal fluids, the frequency of detection of JCV will be increased in patients with AIDS-associated PML compared with healthy controls and HIV-seropositive patients without PML and that higher levels of the JC Virus will correlate with increased immunosuppression.
