This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. We have just begun diffraction studies (F1 line) on the membrane proteins grown in-house by the in meso method. Measurements were made during our first trip to CHESS under the auspices of this grant in September, 2003. Data were collected using cryocooled microcrystals of maximum dimension 40 micron. Best results to date are diffraction to 1.8 ¿ for bacteriorhodopsin (bR) and 2.8 ¿ for the light harvesting complex II (Lh2) protein. Structures of bR have been solved using molecular replacement. Diffraction data statistics for bR/Lh2 are as follows: Space group: P63 / P212121; Resolution limits (¿): 40 - 1.85/50 - 2.8; Unique reflections: 20,214/26,790; Completeness (%): 96.7/98.6; I/sigma: 29.9/13.3; Rsym (%): 7.0/9.4.
