This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. Cosumption of phytoestrogens are suspected to have great impacts on prostate cancer risk.Using a TRansgenic Adenocarcinoma of Mouse Prostate (TRAMP)model, we aim to study the chemopreventive effects of dietary phytoestrogens on prostate cancer progression.We hypothesis that phytoestrogens act through an estrogen receptor (ER)mediated pathway in vivo.We are able to address this ER dependency using the ER deficient mice (ER-alpha KO,ER-beta KO,ER-alpha & beta KO)expressing the TRAMP transgene.The endpoints being measured in this study are:prostate tumor progression;expression of androgen receptor,ER - alpha and ER-beta;differetial methylation of prostate tumor related genes and involvemen of signaling pathways.Understanding mechanisms of chemopreventive effects of phytoestrogens will contribute to prevention and therapeutics of human prosate
