This subproject is one of many research subprojects utilizing the
resources provided by a Center grant funded by NIH/NCRR. The subproject and
investigator (PI) may have received primary funding from another NIH source,
and thus could be represented in other CRISP entries. The institution listed is
for the Center, which is not necessarily the institution for the investigator.
Technical Research Project 3, Product Terminal Isotope Coding (PROTIC), will focus on the
proteomic characterization of the end products of proteolysis. The ability to identify all products, and
then infer the initial substrates, for a protease is essentially the "holy grail" of proteolysis. This
capability would transform the field of cell biology. The significance of the task is matched by its difficulty. We propose to develop an entirely novel chemical strategy for Product Terminal Isotope Coding (PROTIC), the isotope tagging of all N-termini in biological samples. This tagging method will allow the identification and relative
quantification of substrate/product profiles in complex proteomes
