This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The goal of the proposed study is to understand the genetic variation leading to differences in chronic asthma severity between asthmatic individuals. In this proposal we will generate a list of 20 naturally occurring polymorphic variants in asthma-associated genes determined by a literature search. In parallel, we will conduct a cohort study of 1,500 asthmatic children representative of multiple ethnic backgrounds, with equal numbers of males and females. These patients will be genotyped and interviewed to assess chronic asthma severity, as well as medication, medication intervention, and environmental exposure history. This "clinical" data will then be correlated with the polymorphism data to identify those naturally occurring variants, which show a statistically significant association with chronic asthma severity while controlling for medication, medical intervention, and environmental exposure history.
