Real time (RT) polymerase chain reaction (PCR), also known as quantitative PCR (Q-PCR), is a method that amplifies fragments of nucleic acids of known nucleotide sequence and is able to quantify the often very small amounts of this nucleic acid in the original sample. Several aspects of the research pursued in my laboratory (currently funded by NSERC) involve the characterization of the genes encoding proteins that are involved in the cell differentiation process (spermatogenesis) that leads to the formation of male sperm cells (spermatozoa). We are also very interested in the characterization of the proteins that interact with sperm immediately after fertilization of the egg and their gene expression. Therefore, much of our current work involves the analysis and characterization of genes from different vertebrate and invertebrate organisms and the timing and amount of their expression using Q-PCR. In addition to understanding the basic molecular mechanisms involved in spermatogenesis and early embryogenesis, this research has important implications for infertility. In our lab we have been using the PCR technique extensively in the past but the current needs of our research require the accurate quantification of the reaction. To this end, the availability of a Q-PCR machine is imperative. In addition to its contribution to the scientific relevance of the project pursued, this piece of equipment is very important for first-hand training of students and other personnel in the lab. Q-PCR is a widespread current technique used in clinical analysis at hospitals across the country.
