In modern biochemistry and molecular biology laboratories, numerous DNA, RNA and protein manipulations revolve around gel-based electrophoresis separation of these macromolecules. For more than 20 years now, CCD imaging and informatized record keeping have superseded the use of silver nitrate film based technologies. As this technology is quickly evolving, new imaging capabilities are introduced on a regular basis. CCD capture is the best technology to capture DNA and RNA gel data produced from the emission of light after fluorescent excitation of the nucleic acid stained with various intercalating dyes. This can also be applied to proteins and specific stains have been developed to detect, proteins, phosphoporteins, or glycoproteins. With high-resolution cameras, CCD capture provides publication quality results and with increased sensitivity, application such as chemiluminescene is now possible. Combined with epiluminescence and filter sorting of the desired wavelength, multiplexing is now possible and comparative analysis of, for example, 2D electrophoresis gel analysis and spot quantification can be performed on such gel documentation systems with high resolution. The equipment proposed here is primarily intended to cover our increasing needs in: 1) DNA gel analysis and quantification; 2) chemiluminescence detection from protein (Western) blots; 3) fluorescence applications targeted at comparative and differential protein expression analysis.
