In this proposal we request an upgrade to replace three 10-14year-old calcium and pH imaging systems that have become practically unusable due to failure of the CCD chips and problems with the aging software. The requested upgrade will make use of our microscopes and associated equipment that cannot currently function effectively, by forming a single new outstanding system for time lapse and imaging changes in ions in live cells. This upgrade will vastly improve sensitivity to fluorescent dyes and at the same time enhance the resolution and quality of images captured. The new camera and software will significantly improve ease of use by our graduate students and associates thereby increasing both user-time efficiency and training.
