Homebrewing and craft brewing represent an increasingly large portion of the beer industry in Canada and internationally. As a result of the increase in small scale, custom, and craft brewing there is an increased demand for both mixed yeast cultures and characterization of strains within mixed cultures associated with particular styles and flavours. This project will profile different mixed cultures of yeast through genomic DNA sequencing to identify genes and genotypes associated with particular flavours and properties within mixed cultures, along with properties that allow for stable growth and predictable composition. Additionally, the project will develop a sequencing-based methodology to track the long-term stability of mixed yeast cultures as a quality control step to ensure that mixed culture yeast products are consistent over time. Sequencing will be done using Illumina and Oxford Nanopore Technology (ONT) sequencing. This project will assess ONT sequencing as a possible in-house resource for microorganism sequencing, potentially providing an affordable long-read sequencing platform with a rapid turnaround time and a much lower capital cost than other sequencing technologies.
