From a multidisciplinary approach, we will characterize the physicochemical interactions between protein and phenolic ligands, which are playing a major role in many biological processes. These key issues have to be addressed to better understand the biological responses of some natural ligands such as polyphenols. Accumulating data on the protective effects of natural polyphenolic compounds in the progression of neurodegenerative diseases (NDs) has been reported. In this research, we propose a new mechanism to delay the self-association of the protein amyloid-beta (Aß), which could potentially have a positive impact on various biological processes. The amyloid-beta peptide is an endogenous compound which is involved in several pathologies. To test this mechanism, we will combine different multidisciplinary models from theoretical (molecular modelling), experimental (QCMD, real-time nanoparticle tracking) methods and cellular models to better understand the physicochemical interactions between different molecules and their activities.#(cr)#(lf)À partir d'une approche multidisciplinaire, nous caractériserons les interactions physico-chimiques entre les protéines et les ligands phénoliques, qui jouent un rôle majeur dans de nombreux processus biologiques. Ces questions clés doivent être abordées pour mieux comprendre les réponses biologiques de certains ligands naturels tels que les polyphénols. De nombreuses données montrent un effet préventif des composés polyphénoliques naturels dans la progression de maladies neurodégénératives (MN). Dans cette recherche, nous avons proposé un nouveau mécanisme pour retarder l'auto-association de la protéine amyloïde-bêta (Aß), qui pourrait potentiellement avoir une incidence positive sur divers processus biologiques. Le peptide amyloïde-bêta est un composé endogène qui est impliqué dans plusieurs pathologies. Pour cela, nous allons combiner différents modèles multidisciplinaires issus des méthodes théoriques (modélisation moléculaire), expérimentales (QCMD, visualisation des nanoparticules en temps réels) et des modèles cellulaires pour mieux comprendre les interactions physico-chimiques entre différentes molécules et leurs activités.
