牡丹是重要的观赏作物,周年供应主要依靠促成栽培,而花芽休眠解除是促成栽培的关键技术环节。申请者前期工作发现,脱叶和GA3处理均可诱导牡丹花芽休眠解除,且存在加性效应,而糖信号在这一过程中起重要媒介作用。本研究以前期筛选得到的易调控品种‘秋发一号’为对象,明确促成栽培条件下糖信号调控对花芽形态、生理代谢及激素平衡的影响;克隆并研究HXK、SnRK1、TPS1等糖信号调控关键基因的时空表达特性及与GA、ABA等途径的互作,分析潜在作用机制;筛选部分重要基因,通过原位杂交及转基因互补验证等进一步明确其功能;最后选取4-5个关键时间节点,通过转录组测序,以“糖信号-激素互作”为主线,对不同代谢通路进行系统分析,并最终构建糖信号调节牡丹花芽休眠解除的网络调控模型。本研究可为深入了解牡丹花芽休眠解除机制,并为开发更为简易的、多品种适用的牡丹促成栽培技术提供理论基础。

Tree peony is an important ornamental plant, forcing culture meets its annual supply, and bud dormancy release is the key step for this method. We previously found that both defoliation and GA3 application induced tree peony bud dormancy release with additive effect, and sugar signaling played an important role as mediator in this process. In this study, the tree peony cultivar ‘Qiu Fa No. 1’, which has good shallow dormancy character, is chosen as the material. We plan to determine the effect of sugar signaling regulation on the morphological and endogenous metabolic changes as well as hormone balance of tree peony under forcing culture condition; clone and study the transcriptional expression of some key genes in sugar signaling such as HXK, SnRK1 and TPS1, and their crosstalk with GA- and ABA-pathway, and analyze their possible regulating mechanism; select some important genes from them, and confirm their function by in situ hybridization and gene complementarity test; at last, we will systematically analyze different metabolic pathways through transcriptome sequencing, mainly based on the sugar-hormone crosstalk, and finally construct the network model of sugar signaling regulated bud dormancy release. This study may provide a theoretical basis for better understanding the mechanism of bud dormancy release and developing more simple and multi-cultivar suitable forcing culture techniques of tree peony.
