根肿病严重威胁大白菜生产,因此抗根肿病研究成为现今重要课题。自噬在植物抵抗病原物侵染中起重要作用,但其与根肿病抗性关系尚未见报道。申请者前期筛选到一个根肿菌侵染后在大白菜根系中显著上调表达的自噬蛋白BraATG6,并发现拟南芥同源缺失突变体atg6中根肿菌的侵染进程明显加快,说明BraATG6可能具有阻碍根肿菌浸染的作用;网站预测BraATG6可与几个其它自噬蛋白互作,在BraATG6基因启动子上发现有WRKY转录因子结合域,在受根肿菌侵染的大白菜根系中也观察到了自噬现象。由此,推测BraATG6可能受WRKY调控,并与其他自噬蛋白互作引起自噬来抵抗根肿菌侵染。本研究拟进一步开展转基因功能验证、BraATG6蛋白与其它自噬蛋白互作关系验证、以及BraATG6表达是否受WRKY调控等研究,摸清BraATG6在大白菜抵抗根肿菌侵染中的响应机制,为利用自噬抑制根肿菌侵染、防控根肿病发生提供依据。
英文摘要
Clubroot significantly threaten the production of Cruciferous crops,such as Chinese cabbage (Brassica rapa L. ssp. pekinensis), so the research of resistanting to the infection of Plasmodiophora Brassicae is very important. Autophagy plays a key role in the progress of plant defensing to pathogen. In previous research, we obtained a significantly up-regulated autophagy protein, BraATG6, in Chinese cabbage roots inoculated by Plasmodiophora brassicae. The infection process of Plasmodiophora brassicae was significantly faster in Arabidopsis atg6 mutant than in Col-0. These results suggested that BraATG6 may play a key role in preventing the infection of Plasmodiophora brassicae. BraATG6 was predicted to interact with several other autophagy proteins by web software,WRKY transcription factor (TR) binding domain was found on the promoter of BraATG6. Otherwise, autophagy was observed in Chinese cabbage roots inoculated by Plasmodiophora brassicae. So we guess BraATG6 being regulated by WRKY and interacted with several other autophagy proteins to induce autophagy to resistant the infection of Plasmodiophora brassicae. In this research, the function of BraATG6 will be analysised through the genetic engineering means, the interaction of BraATG6 and other autophagy proteins will be verified, and it will be detected that the regulation role of transcription factor WRKY to the expression of BraATG6. Together with above analysis, it will be revealed that the mechanism of BraATG6 responding to the progress of Chinese cabbage resistanting to the infection of Plasmodiophora brassicae. These results will help us provide clues by using autophagy to prevent the infection of Plasmodiophora brassicae and then to control the clubroot.
