肥胖与诸多重大疾病密切相关,最近流行病学研究表明抗生素可诱发肥胖。随着环境抗生素污染水平不断上升,阐明其诱胖效应与潜在作用机理是亟待解决的关键科学问题。本项目在前期研究基础上,拟选择国内外常用的肥胖模式生物秀丽线虫为对象,围绕脂代谢紊乱、胰岛素抗性和饱腹感反馈迟钝/饮食过量等方面及其相互作用,研究环境中典型抗生素诱发肥胖效应与潜在机制;选择与肥胖发生关系密切的肠道菌代表——大肠杆菌,从肠道菌脂代谢与宿主秀丽线虫脂代谢的相互关系上,研究抗生素暴露下肠道菌脂代谢变化对宿主脂代谢与调控的影响及其肥胖效应,探索抗生素诱发宿主肥胖效应中肠道菌群的介导作用,从全新的视角深入揭示环境抗生素诱胖机制;研究抗生素诱发亲代肥胖后对后代肥胖、生长发育、运动行为、生殖与寿命等健康影响,理解抗生素诱发肥胖的长期效应。本项目旨在阐释抗生素诱发肥胖效应与潜在机理,为环境抗生素的长期健康风险评价提供科学依据与方法。
英文摘要
Obesity is related with serious health problems. Recent epidemiological studies demonstrated that antibiotics can provoke obesity. The pollution levels of environmental antibiotics are increasing, and their obesogenic effects and potential mechanisms are one key scientific question to be answered. On the basis of our previous studies, we will perform the present proposal in the following way. First, we will choose the model organism of obesity Caenorhabditis elegans as the target, and study disturbances on lipid metabolism, insulin resistance, delay of satiety and over-eating, and the connections among them to elucidate the potential obesogenic mechanisms of antibiotics. Then, we will choose Escherichia coli to represent microbiota that are quite related with obesity. We focus on the connection of lipid metabolism between microbiota and the host, and will study how the changes in lipid metabolism of microbiota influence the host lipid metabolism and its regulation, and also the obesogenic effects. Such studies will elucidate the mediation mechanisms of microbiota in host obesity, providing new information to explain the obesogenic mechanisms of environmental antibiotics. Moreover, after the obesogenic effects of the antibiotics on the pregnant mothers, we will further study the effects on the health of the offspring including obesity, development, locomotion behavior, reproduction and lifespan to explore the long-term influences of obesity. The achievements of the present study will answer the obesogenic effects and mechanisms of environmental antibiotics, and provide scientific supports and methods for the long-term health risk assessment on environmental antibiotics.
