本项目以黑线仓鼠为材料,采用不同温度、光周期和食物丰度三个主要随季节变化的环境因子处理黑线仓鼠,探讨调控黑线仓鼠下丘脑PACAP表达的主要环境因子;通过外源PACAP注射,研究PACAP调控GnRH表达的遗传效应;通过转录组测序、GO功能和KEGG Pathway分析、噬菌体展示和酵母杂交技术,研究主要环境因子-PACAP-GnRH调控黑线仓鼠繁殖活动的分子路径;采用实时定量PCR技术、免疫组化等方法研究主要环境因子-PACAP-GnRH路径中各因子的表达量;用酶联免疫法检测血液FSH和LH的浓度,并统计卵巢卵泡数量;利用SPSS软件分析各因子对黑线仓鼠繁殖活动的调控效应。该研究有利于掌握主要环境因子-PACAP-GnRH路径中调控黑线仓鼠繁殖活动的主效因子,为合理控制农业害鼠繁殖活动提供理想靶点,为农业害鼠综合治理措施的制定提供新的思路和方法。

In this study, the striped hamsters (Cricetulus barabensis) were treated with different levels of temperature, photoperiod and the abundance of food that significantly change with the seasons to explore the main environmental factor that regulates the expression of PACAP in the hypothalamus of the striped hamsters; The genetic effects of PACAP on the expression of GnRH in the hypothalamus of the striped hamsters were studied by the injection of exogenous PACAP; By means of the transcriptome sequencing, GO function and KEGG Pathway analysis, the phage displaying technology and the yeast hybridization technology, the molecular mechanism of the main environmental factors-PACAP-GnRH pathway on regulating the reproductive activity of the striped hamsters will be studied; The expression level of the factors in the main environmental factor-PACAP-GnRH pathway will be examined by Real-time quantitative PCR, Immunohistochemistry and In situ hybridization technologies; The concentrations of FSH and LH in blood were detected by enzyme-linked immunosorbent assay (ELISA), and the number of follicles in ovarian was counted; The regulative effects of the factors in the main environmental factor-PACAP-GnRH pathway on reproductive activities in the striped hamsters were also analyzed by SPSS software. This research is beneficial to understand the main factors regulating the reproduction of the striped hamsters in the main environmental factor-PACAP-GnRH pathway, which provides ideal targets for controlling the reproduction of the agricultural rodents, and applies new ideas and methods for making comprehensive measures to control the abundance of the agricultural rodents.
