源于远缘杂交形成的新型异源三倍体鱼—湘云鲫2号(3N=150)具备生长速度快,存活率高和抗逆性强的杂种优势,且与母本红鲫(2N=100,♀)不仅在遗传组成、个体形态及生理学上存在差异,而且外周血组成及理化特性上也存在差异,这为研究硬骨鱼免疫应答和代谢特征提供了良好的动物模型。然而,有关湘云鲫2号与母本红鲫外周血的免疫应答和病理代谢的差异尚不清楚。本项目拟以转录组和代谢组关联分析嗜水气单胞菌感染后湘云鲫2号及其母本红鲫外周血生物标记代谢物的调控特点及外周血单个核细胞(PBMC)免疫应答差异。在此基础上,结合实时荧光定量PCR、蛋白印迹等分子生物学方法,比较不同倍性鲫外周血的免疫力差异,分析PBMC免疫应答调控特点,发掘外周血生物标记代谢物特征及调控差异,以期为远缘杂交形成的异源三倍体鱼的抗病机制研究提供重要的数据。本研究在鱼类抗病育种理论研究和应用研究上具有重要的指导意义。

The new type of allotriploid fish-Xiangyun crucian carp No. 2 (3N=150) has the advantages of fast growth, high survival rate and strong resistance to stress. Moreover, there are differences not only in genetic composition, individual morphology and physiology, but also in peripheral blood composition and physicochemical characteristics between Xiangyun crucian carp No. 2 and its female parent, red crucian carp (2N=100, ♀), which can be used as an animal model to study the immune response and metabolism in teleost. However, the difference of immune response and pathological metabolism between Xiangyun crucian carp No. 2 and red crucian carp remains unclear. This project aims to analyze the regulation characteristics of biomarker metabolites in peripheral blood of Xiangyun crucian carp No. 2 and its female parent red crucian carp and the difference of PBMC immune response after Aeromonas hydrophila infection. On this basis, we compared the immunity differences of peripheral blood between Xiangyun crucian carp No. 2 and red crucian carp, analyzed the characteristics of PBMC immune response, and explored the characteristics and regulation differences of peripheral blood biomarker metabolites, in order to provide important information for the study of pathological metabolism and disease resistance mechanism of allotriploid fish. This study has an important guiding role in the theoretical research and application research of fish disease resistance breeding.
