兽药多残留的自动化、高灵敏度、高通量免疫分析技术是动物源性食品安全研究的热点和难点;相对于医学检验中全自动化学发光免疫分析技术,当前兽药残留免疫分析方法存在自动化程度低、灵敏度差、检测通量低等缺陷。本研究拟选取磺胺类和喹诺酮类兽药作为代表性分析物,通过软光刻技术构建液滴微流控芯片,制备油相包裹两类兽药抗原与抗体免疫反应体系的纳升级水相液滴作为独立免疫分析单元,评价荧光编码微球与荧光共振能量转移分子探针在兽药残留免疫分析中的应用性能,筛选适用于液滴微流控芯片的双标记荧光编码探针,开展双色流式荧光免疫分析方法研究;优化微流控芯片通道设计、液滴大小、液滴内抗原与抗体加载量等参数,实现自动化、高灵敏度、高通量同步检测磺胺类和喹诺酮类兽药多残留。基于液滴微流控技术构建双色流式荧光免疫分析方法,将为畜禽产品中兽药多残留新型免疫分析方法的设计、改进和发展提供指导信息和新思路,具有重要理论意义和应用价值。

The multiple veterinary drug residues in foods of animal origin could pose hazard on human health. To develop multi-analyte immunoassays for simultaneously detecting multiple veterinary drug residues are in urgent need to solve food safety problems in China. It is also a world-wide food safety concern to develop automatic, higher sensitive and through-put multiplex immunoassays for veterinary drug residue analysis in foods of animal origin. Compared with the automatic chemiluminescent immunoassay in laboratory medicine and clinical sciences, traditional immunoassay formats still have the disadvantages of low automaticity, bad sensitivity and low-throughput for veterinary drug residue analysis. In order to improve the assay performance, we want to combine the droplet-based microfluidics techniques with the flow cytometric immunoassays to detect multiple veterinary drug residues in foods of animal origin. In this study, the sulfonamides and fluoroquinolones are selected as model analytes, and we will use the photolithography technology to produce the droplet-based microfluidic chips. The micro-droplets are produced and utilized as immuno-reaction carriers by encapsulation the two kinds of antigen-antibody complexes with oils. Besides, two kinds of fluorescence microspheres with different colors, and two pairs of fluorescence resonance energy transfer donors and receptors are used as labels to identify the sulfonamides and fluoroquinolones. The related fluorescent intensities are recorded as signals for calculation the target concentration of the sulfonamides and fluoroquinolones residues in foods of animal origin. After combining the droplet-based microfluidics with the flow cytometry technology, the developed multiplex flow cytometric immunoassays have the advantages of higher automaticity, sensitivity and high-throughput. And it could enrich the existing immunological analytical methods for veterinary drug residue analysis in foods of animal origin. The implement of this project will provide useful information on the design and improvement of the new immunoassay formats for multiple veterinary drug residue analysis.
