柑橘木虱是世界柑橘产业毁灭性病害-柑橘黄龙病(HLB)的主要虫媒,HLB病原菌CLas不能人工培养,却能专性侵染柑橘木虱并在其体内多个组织器官中繁殖生长。项目组前期研究发现CLas侵染柑橘木虱中肠能够引起宿主细胞内吞和蛋白酶体等途径相关因子变化。本项目拟采用CLas侵染柑橘木虱中肠转录组与蛋白质组联合分析、qPCR和Western-blot验证为基础,综合运用特异性抑制剂和关键基因RNAi等技术分别对网格蛋白介导的内吞途径和泛素-蛋白酶体系统(UPS)等开展分段式阻断或沉默研究,结合CLas病菌细胞内荧光原位杂交(FISH)定位、亚细胞组分特异性染色与显微观测技术,对CLas入侵柑橘木虱中肠的方式、胞内转运及其免疫逃避途径开展研究,确定CLas侵染柑橘木虱的关键控制靶点,为进一步研究柑橘黄龙病防治新策略、控制媒介昆虫的病原传播提供理论依据。

Citrus psyllid(Diaphorina citri,Hemiptera) is the major vector that transmits Huanglongbing(HLB), which is a devastating disease of citrus caused by ‘Candidatus Liberibacter asiaticus’ (CLas). The CLas still be not artificial cultivated, but it can specifity infect citrus psyllid and reproductive growth in its multiple tissues and organs. We have found that CLas invasion can cause changes of endocytosis and proteasome pathway and related factors in host cells from midgut of citrus psyllid in previous studies. The project intends to adopt quantitative proteome combined with transcriptome study and qPCR and Western-blot verification as preliminary studies. Integrated apply specific inhibitors and RNAi for key genes or protein, in order to segmented blockade or silencing clathrin-mediated endocytic pathway and ubiquitin-proteasome system (UPS), respectively. Assist to fluorescence in situ hybridizations (FISH) for the localization of CLas and subcellular component specific staining in epithelial cells of midgut combined with microscopic observation technique. Our perspective will reveal the ways of CLas invading, intracellular transport and immune evasion pathways in the epithelial cells from midgut of citrus psyllid. Identify key control targets for CLas infection of citrus psyllids. The results will conduce to further study the new strategy of citrus Huanglongbing prevention and treatment, and also benefit to support for studying on interaction pathogen between vector-borne and control the transmission of the pathogen.
