我们研究了Lenti病毒介导的基因治疗Krabbe病的效果。我们通过大规模收获和超速离心法成功地制备了含GALC、Krabbe病表达基因缺陷酶的高滴度Lenti病毒，并注射给新生Twitcher小鼠，有效地改善了Krabbe病模型的营养和寿命。采用L-环丝氨酸基因治疗和底物还原治疗相结合的方法能更有效地改善Twitcher小鼠的表型。
英文摘要
We investigated the effect of lenti-virus mediated gene therapy for Krabbe disease. We succeeded in preparing high titer lenti-virus including GALC, deficient enzyme in Krabbe disease expressing gene by large scale harvesting and ultracentrifuge, and injected for neonatal twitcher mouse, model for Krabbe disease resulted in efficient improvement in nutrition and life span. And we had more efficient improvement in the phenotype of twitcher mice by the combination of gene therapy and substrate reduction therapy using L-cycloserine.
