This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. We will expand our ongoing efforts in the development of microsphere-based assays to develop a new approach to the genetic analysis of environmental samples that will enable sensitive and rapid detection of pathogens with a high degree of confidence and a low false alarm rate. Specifically, we will develop multiplex microsphere-based assay kits to provide rapid point detection of pathogen-specific PCR products amplified from environmental samples, allowing screening for a panel of organisms in a single test. Because flow cytometry is the analysis platform, sample preparation is simple, requiring no wash steps, and analysis is rapid, less than one minute. The new assays will be compatible with commercial laboratory flow cytometers as well as smaller fieldable instruments, including the Autonomous Pathogen Detector (APD) being developed as part of the DOE CBNP effort. These new tools will allow flexible design to provide rapid analysis in a variety of response scenarios.
