歯肉溝滲出液(GCF)中のコラゲナーゼ活性、線維芽細胞型コラゲナーゼ(MMP-1)及び好中球型コラゲナーゼ(MMP-8)、それらのMMPのインヒビターであるTissue inhibitor of MMPs(TIMP)-1及びTIMP-2のレベルと歯周組織の炎症あるいは組織破壊のレベルとの関連性について検索した。研究には歯周炎患者(11名)、歯肉炎患者(10名)、健常者(10名)より採取したGCFを供し、TIMP-1及びTIMP-2はELISAにより、また、コラゲナーゼ活性 [活性型及びAPMA(p-aminophenyl mercuric acetate)活性型]については酵素分解物をSDS-PAGE電気泳動後、画像解析を応用して測定した。その結果、GCF中のTIMP-及びコラゲナーゼ活性のレベルは健常者に比較し歯肉炎及び歯周炎患者群で有意に高かった[TIMP-1(ng):健常者3.49±2.91、歯肉炎11.18±6.30、歯周炎13.68±9.64、活性型コラゲナーゼ活性(Units):健常者0.226±0.181、歯肉炎0.360±0.387、歯周炎0.623±0.409、APMA活性型コラゲナーゼ活性(Units):健常者0.318±0.205、歯肉炎0.664±0.372、歯周炎0.880±0.403]。TIMP-2は全被検者の29.0%のみで検出できた(2.06ng)。また、ウェスタンブロットの結果、MMP-8が歯肉炎及び歯周炎患者群GCF中の主要なコラゲナーゼであることが確認された。さらに、今回得られたデータをいくつかの臨床指数と共に主成分分析という統計学的手法にかけることにより、全体を87%説明できる3つの主成分が抽出され、コラゲナーゼ活性(特に活性型)が歯周炎の病態を特徴付ける極めて重要な因子であり、診断あるいは治療上有意義なマーカーである可能性が示唆された。

Tissue inhibitor of MMP (TIMP)-1 and TIMP-2 in dental fissure exudate (GCF), tissue inhibitor of MMPs(MMP-1) and tissue inhibitor of MMPs(MMP-8) in dental fissure exudate (GCF), tissue inhibitor of MMPs(MMPs-1) and tissue inhibitor of MMPs(MMP-2) in dental fissure exudate (GCF), tissue inhibitor of MMPs(MMPs-1) and tissue inhibitor of MMPs(MMP-2) in dental fissure exudate (GCF). In the study, GCF, TIMP-1 and TIMP-2 were detected in 11 patients with periodontitis, 10 patients with odontitis and 10 healthy subjects by ELISA, SDS-PAGE and image analysis. Results: TIMP-1(ng) in GCF was significantly higher in healthy patients than in periodontitis and periodontitis. 3.49±2.91 in normal group, 11.18±6.30 in periodontitis, 13.68±9.64 in active group, 3.49±2.91 in healthy group, 11.18±6.30 in active group, 13.68±9.64 in active group.(Units): Normal 0.226±0.181, Dentitis 0.360±0.387, Periodontitis 0.623±0.409, APMA Active Type (Units): normal 0.318±0.205, periodontitis 0.664±0.372, periodontitis 0.880±0.403]. TIMP-2 was detected in 29.0% of all detected samples (2.06ng). The results of MMP-8 in patients with periodontitis and periodontitis confirmed that MMP-8 was a major factor in GCF. The clinical index of periodontitis was analyzed by principal component analysis (PCA). The statistical method was used to extract the principal component of periodontitis. The activity of periodontitis was analyzed by PCA. The probability of periodontitis was analyzed by PCA.
