项目摘要

1)消化器癌における新しい癌関連遺伝子異常の検索本研究の目的である癌関連遺伝子異常と患者予後との関連をみるため、消化器癌におけるcell cycIe関連遺伝子に関して研究を進めた。特に新しい癌関連kinaseであるaurora2(AUR2)に注目し、まずAUR2のprimerを作製後、COLO205,HT29,Helaの細胞を用いて条件を設定し、RT-PCR法によるAUR2発現の検索システムを確立した。2)ヒト消化管癌におけるAUR2発現とリンパ節転移との関連AUR2 mRNA発現は食道癌51%(27/53)、胃癌53%(38/72)、大腸癌47%(36/76)と各消化管癌の約半数に認め、その多<はT/N比が10を超え腫瘍特異的に発現していた。臨床病理学的にAUR2陽性群と陰性群を比較すると、胃癌ではリンパ節転移率が陽性群71%(27/38)、陰性群38%(13/34)で、陽性群は陰性群に比べ有意にリンパ節転移が多かった(p<0.01)。しかし病期・組織型など他の因子は両群間に差はなかった。大腸癌でもリンパ節転移率は陽性群64%(23/36)、陰性群18%(7/40)で、陽性群は有意にリンパ節転移が多く(p<0.01)、リンパ管侵襲も陽性群69%、陰性群30%で、陽性群に有意に多く認めた(p<0.01)。しかし食道癌においてはリンパ節転移は陽性群86%、陰性群76%で両群間に差はなく、所属リンパ節群別にみても差はなかった。予後では大腸癌の3年無再発生存率はAUR2発現陽性群67%、陰性群89%であり、陽性群は有意に予後不良であった(p<0.05)。胃癌でも大腸癌と同様に陽性群の予後不良の傾向がみられたが、食道癌では両群間に差はなかった。この結果よリAUR2の発現は胃癌・大腸癌の悪性度評価、特にリンパ節転移の新しい指標になり得ると考えられた。

1)The purpose of this study is to further study the relationship between cell cycIe related gene abnormalities and patients with digestive cancer. In particular, the new cancer-associated kinase aurora2(AUR2) attracted attention. After the AUR2 primer was produced, the conditions for using COLO205, HT29, and Hela cells were set, and the model for the discovery of AUR2 by RT-PCR was established. 2) AUR2 mRNA expression in digestive tract cancer was associated with tumor metastasis in 51%(27/53) of esophageal cancer, 53%(38/72) of gastric cancer and 47%(36/76) of colorectal cancer. In clinical pathology, AUR2 positive group and negative group were compared. In gastric cancer, 71%(27/38) of positive group and 38%(13/34) of negative group were shifted. The disease period, tissue type, and other factors are different from each other. In colorectal cancer, the rate of tumor metastasis was 64%(23/36) in positive group, 18%(7/40) in negative group, 69% in positive group, 30% in negative group and 70% in positive group. 86% of esophageal cancer patients were positive, 76% were negative, and 86% were negative. The 3-year recurrence-free survival rate of colorectal cancer was 67% in AUR2 positive group, 89% in AUR2 negative group, and 89% in AUR2 positive group (p<0.05).
